Background: Interleukin-1 beta (IL-1β), also known as catabolin, is a cytokine protein encoded by the IL1B gene in humans. The precursor of IL-1β is cleaved by caspase 1 (interleukin 1 beta convertase), and a cytosolic thiol protease further processes the product into mature IL-1β. IL1β and IL1α are closely related polypeptides sharing approximately 21% amino acid identity in humans. They are synthesized by various cells in response to inflammatory stimuli, infections, or microbial endotoxins. Despite being regulated independently, both IL1α and IL1β bind to the same receptor and exert identical biological effects.

Belonging to the interleukin 1 cytokine family, IL-1β is primarily produced by activated macrophages as a proprotein. Caspase 1 (CASP1/ICE) enzymatically converts it into its active form. This cytokine plays a crucial role as a mediator of the inflammatory response and participates in diverse cellular activities such as proliferation, differentiation, and apoptosis. Notably, IL-1β induces the expression of cyclooxygenase-2 (PTGS2/COX2) in the central nervous system (CNS), contributing to inflammatory pain hypersensitivity. The IL1B gene, along with eight other interleukin 1 family genes, is clustered on chromosome 2.
