Background: Cystatin C belongs to family 2 of the Cystatin superfamily and plays roles in various physiological processes including tumor invasion, metastasis, inflammation, and certain neurological disorders. It functions by inhibiting numerous cysteine proteases such as papain, as well as cathepsins B, H, K, L, and S. Found widely across human tissues and body fluids, Cystatin C is crucial in regulating protease activity.

A single point mutation in the gene encoding the 120-amino acid mature Cystatin C (Leu68 to Gln) results in a hereditary form of amyloid angiopathy. This variant protein accumulates in cerebral arteries, leading to fatal cerebral hemorrhage. Beyond this pathology, Cystatin C holds promise in clinical settings, serving as a reliable marker for assessing glomerular filtration rate.
