Background: | Neural cell adhesion molecule 1 (NCAM-1) is a versatile member of the Ig superfamily, playing crucial roles in various cellular interactions. Belonging to a group of membrane-bound glycoproteins, it engages in Ca++-independent cell-matrix interactions as well as homophilic or heterophilic cell-cell adhesions. NCAM-1 exhibits specific binding capabilities to heparan sulfate proteoglycans, agrin, and several chondroitin sulfate proteoglycans including neurocan and phosphocan. Human NCAM-1 manifests in three principal forms resulting from alternate splicing: NCAM-120/NCAM-1 (761 amino acids [aa]), NCAM-140 (848 aa), and NCAM-180 (1120 aa), where NCAM-120 is GPI-linked, while NCAM‑140 and NCAM-180 are type I transmembrane glycoproteins. Extensive alternate splicing and possible extracellular proteolytic processing further diversify all three forms. Synthesized as a 761 aa preproprecursor with a 19 aa signal sequence, NCAM-1 consists of a 722 aa GPI-linked mature region and a 20 aa C-terminal prosegment, featuring five C-2 type Ig-like domains and two fibronectin type-III domains. Human and mouse NCAM-1 share 93% aa identity, with NCAM-1 being highly sialylated. Polysialylation of NCAM-1 diminishes its adhesive properties while enhancing neurite outgrowth promotion. Notably, the adult brain displays reduced sialylation of NCAM-1 compared to earlier developmental stages, yet regions exhibiting heightened neuronal plasticity maintain polysialylation-NCAM-1, indicating its involvement in regenerative processes and synaptic plasticity. |
