Background: Ubiquitin-conjugating enzyme E2 D3, encoded by the UBE2D3 gene in humans, is a member of the ubiquitin-conjugating enzyme family. These enzymes, also known as E2 enzymes or ubiquitin-carrier enzymes, play a crucial role in the ubiquitination process's second step. Initially, E1 activates ubiquitin by attaching it covalently to its active site cysteine residue. The activated ubiquitin is subsequently transferred to a cysteine residue on an E2 enzyme. E2 enzymes then interact with E3 ligases through a structurally conserved binding region. This ubiquitination process modifies proteins, thereby regulating protein degradation.

UBE2D3 is the human counterpart of the yeast UBC4/5 family and is involved in numerous regulatory functions linked to inflammation and cancer. It facilitates the degradation of various short-lived regulatory proteins, such as p53 in the presence of E6/E6-AP or MDM2, c-Fos, IκBα, and p105. UBE2D3 shares significant sequence identity, 88% with UbcH5a and 89% with UbcH5b, highlighting its structural and functional similarities to these related E2 enzymes.
