Background: | Ubiquitin-conjugating enzyme E2 L3, encoded by the UBE2L3 gene in humans, is a member of the ubiquitin-conjugating enzyme family. These enzymes, also known as E2 enzymes or ubiquitin-carrier enzymes, play a crucial role in the second step of ubiquitination. Initially, E1 activates ubiquitin by attaching it covalently to its active site cysteine residue. Subsequently, the activated ubiquitin is transferred to a cysteine residue on an E2 enzyme. The E2 molecule then binds to an E3 ligase via a conserved binding region. UBE2L3 specifically interacts with HECT-type and RBR family E3 ubiquitin-protein ligases, including UBOX5, ARIH1, Cbl, UBE3A, and NEDD4. It is down-regulated during the S-phase of the cell cycle, influencing cell cycle progression. Additionally, UBE2L3 regulates the transcriptional activity of nuclear hormone receptors and plays a role in myelopoiesis. |
