Noggin is part of a group of soluble proteins that bind to TGF-β family ligands, regulating their activity by blocking their access to signaling receptors. Originally identified as an antagonist of BMP-4, Noggin is crucial for the proper formation of the head and other dorsal structures. It also modulates the activities of other BMPs such as BMP-2, BMP-7, BMP-13, and BMP-14. In mice, deletion of the noggin gene leads to prenatal death and a recessive phenotype characterized by severely malformed skeletal structures. Conversely, transgenic mice that overexpress noggin in mature osteoblasts exhibit impaired osteoblastic differentiation, reduced bone formation, and severe osteoporosis. Recombinant Murine Noggin is a 46.4 kDa homodimer linked by disulfide bonds, composed of two 206 amino acid polypeptide chains. |
