Y-27632 is a highly potent and selective ATP-competitive inhibitor of Rho-associated kinases (ROCK), exhibiting Ki values of 0.22 µM for ROCK1 and 0.30 µM for ROCK2. In vitro kinase assays reveal that Y-27632 has a 20-fold higher preference for ROCK compared to citron kinase and protein kinase N. Studies indicate that Y-27632 treatment in cells inhibits ROCK-mediated phosphorylation of myosin phosphatase subunit 1 (MYPT1) at Thr853 and Thr696, with a significantly stronger inhibition at Thr853. Additionally, Y-27632 selectively inhibits smooth muscle contraction by preventing Ca2+ sensitization. Further research shows that Y-27632 reduces dissociation-induced apoptosis in cultured human embryonic stem cells over multiple passages and enhances cloning efficiency.
